Derived from roots of 3 week old hydroponically grown unstressed barley cv. Optic plants.   Individual seed of barley were supported and germinated on agar in tubes with drilled out cores to allow roots to grow out into a nutrient solution.  After 7 days at 20C the tubes were placed directly into hydroponics. The nutrient solution consisted of half-strength Hewitt's solution with additional Na2SiO3.  Solutions were vigorously aerated, and replenished weekly with checks on electrical conductivity, pH, volume and O2.  Experiments were conducted in a ventilated glasshouse at 16-24C.

Individual seed of barley were supported and germinated on agar in tubes with drilled out cores to allow roots to grow out into a nutrient solution. After 7 days at 20C the tubes were placed directly into hydroponics. The nutrient solution consisted of half-strength Hewitt's solution with additional Na2SiO3.  Solutions were vigorously aerated, and replenished weekly with checks on electrical conductivity, pH, volume and O2. Experiments were conducted in a ventilated glasshouse at 16-24C. After two weeks in control conditions seedlings were raised above the hydroponics solution so that their roots were in the air for 3 hours daily, after which they were returned. Plants were normally harvested after 2 weeks of treatment.

After 7 days at 20C the tubes were placed directly into hydroponics. The nutrient solution consisted of half-strength Hewitt's solution with additional Na2SiO3.  Solutions were vigorously aerated, and replenished weekly with checks on electrical conductivity, pH, volume and O2. Experiments were conducted in a ventilated glasshouse at 16-24C. After two weeks in control conditions the hydroponics solution was replenished on a weekly basis with normal nutrients minus nitrogen. Plants were normally sampled after two weeks of treatment.

After 7 days at 20C the tubes were placed directly into hydroponics. The nutrient solution consisted of half-strength Hewitt's solution with additional Na2SiO3.  Solutions were vigorously aerated, and replenished weekly with checks on electrical conductivity, pH, volume and O2. Experiments were conducted in a ventilated glasshouse at 16-24C. After two weeks in control conditions salt was added in daily increments to give a final concentration of 150 mol m-3 NaCl. CaCl2 was also added to give a constant ratio of Na+:Ca2+ of 20:1. Plants were normally harvested after 2 weeks of adding the salt.

After 7 days at 20C the tubes were placed directly into hydroponics. The nutrient solution consisted of half-strength Hewitt's solution with additional Na2SiO3.  Solutions were vigorously aerated, and replenished weekly with checks on electrical conductivity, pH, volume and O2. Experiments were conducted in a ventilated glasshouse at 16-24C. After two weeks in control conditions the aeration system was switched off. Plants were normally sampled after two weeks of treatment.

Barley root total RNA was extracted in triplicate from frozen tissue using Trizol reagent (Invitrogen). Total RNA samples were treated with DNaseI and purified through the RNeasy midi columns (Qiagen).
