Barley (Hordeum vulgare L. cv. Graphic) seeds were carefully chosen with similar weight, shape and size. Seeds were placed in Petri dishes and germinated with water at 22ºC in a growth chamber for two days. When radicules could be seen, plantlets were transferred to 1 L pots containing sand as a substrate. During two weeks, plants were grown under glasshouse at the Experimental Fields of the University of Barcelona and irrigated with water in a semi-hydroponic system. Water was changed every two days. After two weeks three modified Höagland (pH 6.0) solutions were applied containing: 1mM P, 5mM K, 1.5 mM Mg, 3.5 mM Ca, 0.1 mM Fe, 0.023 mM B, 0.0045 mM Mn, 0.0003 mM Cu, 0.0015 mM Zn, 0.0001 mM Mo, with either 11 mM NO3-, 11 mM NH4+ or a mixture of both (11 mM NO3- plus NH4+, 1:1). New nutritive solutions were added after 24 h and pH was monitored to avoid any acidification or alkalisation normally produced by NH4+ and NO3- assimilation, respectively. This procedure also prevented NH4+ to be converted to NO3- by micro organisms (Padgett and Leonard, 1993). After 48 h plantlets were sampled, roots were separated from shoots, immediately frozen in liquid N and kept at -80ºC until use. Three individuals (shoots)per nitrogen treatment, were used for the expresion studies.

Total RNA was extracted from entire shoots with Trizol reagent (Invitrogen Life Technologies, Paisley, UK) in three biological replicates and further purified with RNeasy Mini Kit (Quiagen GmbH, Hilden, Germany) following the instructions provided by the supplier. RNA quality and quantity were determined by spectrophotometer and visualization in agarose gel
