Amplification and biotin labeling of cRNA was done using MEGAscript TM High Yield Transcription Kit (Ambion Ltd., Huntingdon, United Kingdom). Briefly, half standard setup including 1.75ml of each Biotin-11-CTP (10 mM) and Biotin-16-UTP (10mM, both from Enzo, Farmingdale, United States), 1.0ml of each T7 ATP (75mM) and T7 GTP (75mM), and 0.75ml each of T7 CTP (75mM) and 0.75ml of T7 UTP (75mM), was used for cRNA synthesis. Finally, the cleanup of labeled cRNA was carried out using RNeasy mini columns (Qiagen, Hilden, Germany).
