The Tn5Prime library preparation method used to analyze these cells is a modification of the Smartseq2 method. It uses a template switch oligo that incorporates a Illumina adapter sequence and a cellular index into cDNA. The cDNA of many cells can then be pooled for PCR. The other Illumina adapter is added by the Tn5 enzyme  loaded only with that one adapter. In this way, Read 1 is anchored to the 5' end of transcripts while Read 2 is distributed randomly throughout the transcripts.
